Abstract


Geobacillus stearothermophilus L-07 was tested for production of extracellular β-mannanase. The enzymes
production was showed by clear zone from around colony after staining with congo red and destaining with
NaCl solution. The enzymes were endohydrolases that catalyze random hydrolysis of β-1,4-mannosidic
linkages in the main chain of galactomannan to mannooligosaccharide, and mannose. The β-mannanase
activity was highest in culture medium, containing 0.65% (w/v) locust bean gum in pH 7.0 after 36 hours of
cultivation at 60oC. However, the highest β-mannanase activity was observed in culture medium after 12
hours of cultivation at 70oC, respectively.
